چکیده

Background: Adrenomedullin (ADM) is a vasoactive peptide hormone found in adrenal medulla, central nervous system, immune cells, and muscle, expressed the uterus placenta. ADM plays important roles angiogenesis, vasodilation, inflammation. Altered levels of have been implicated adverse pregnancy outcomes like fetal growth restriction pre-eclampsia. Animal models can be useful tools for examining pathophysiology. Much what known about has learned from mouse model; however, rat superior model human placentation due to deep hemochorial invasion. Objective: We aim explore role at maternal-placental interface characterize wild type null pregnancy. was localized within site by situ hybridization. used Crispr-Cas9-mediated genome editing create mutant Adm knockout rat. The mutation targeted Exon 2 introduced premature stop codon, resulting 206 bp deletion truncated, nonfunctional protein. Germline transmission confirmed. Heterozygous intercrosses failed produce live rats. heterozygous pairs were bred examine placental growth, development, demise through gestation. placentas/fetuses viable gestation day (gd) 13.5, but all had demised or resorbed gd 16.5. Growth evident 13.5 fetuses. Many conceptus demonstrated fetal-placental hemorrhage infarct. On microscopic evaluation, some deficient sites intrauterine trophoblast giant cells decidual early deficiency disrupts invasion uterine-placental Loss results abnormalities placentation, restriction, death Thus, critical successful favorable outcomes.View Large Image Figure ViewerDownload Hi-res image Download (PPT)

Identification of uterine pacemaker regions at the myometrial-placental interface

Coordinated uterine contractions at the end of gestation are essential for delivering viable offspring in mammals. Contractions are initiated by an electrical signal at the plasma membrane of uterine muscle cells, leading to voltage-dependent calcium entry, and subsequent activation of the intracellular contractile machinery. In contrast to other visceral smooth muscles, it is not known where e...

Trophoblast-uterine interactions at implantation

Implantation of the embryo in the uterus is a critical and complex event and its failure is widely considered an impediment to improved success in assisted reproduction. Depending on whether placentation is invasive or superficial (epitheliochorial), the embryo may interact transiently or undergo a prolonged adhesive interaction with the uterine epithelium. Numerous candidate interactions have ...

Possible roles for folic acid in the regulation of trophoblast invasion and placental development in normal early human pregnancy.

In addition to its role in the prevention of neural tube defects, folic acid has many other physiological functions, including cell proliferation, DNA replication, and antioxidant protection. The aim of this study was to determine the role that folic acid has in regulating placental trophoblast development. Placental explants from placentae at gestational age 7 wk (n = 3) were cultured in folic...

Spontaneous Heterotopic Pregnancy after Four Previous Caesarean Sections: Successful Salpingectomy and Inta-Uterine Term Pregnancy

Heterotopic pregnancy is rare under natural circumstance (conception). We report the first case of heterotopic pregnancy in spontaneous conception with four previous caesarean sections and ruptured tubal ectopic with resultant successful normal intra-uterine pregnancy delivered at term after salpingectomy. A 32-year-old gravida 5 para 4+0 with 3 living children, with 4 previous caesarean sectio...

Integrin switching regulates normal trophoblast invasion.

Cells invade extracellular matrices in a regulated manner at specific times and places during normal development. A dramatic example is trophoblast invasion of the uterine wall. Previous studies have shown that differentiation of trophoblasts to an invasive phenotype is accompanied by temporally and spatially regulated switching of their integrin repertoire. In the first trimester human placent...
